Should my personal essay relate to my major?

I want to major in Biotechnology, but I am writing about a disabled family member and how living with them has shaped me into who I am today. Should my personal essay be about what I want to major in instead? I am doing EC's that relate to my major, should I write about those experiences instead?

I would see how to tie the two together. For example, "seeing my disabled family member struggle and persevere demonstrated grit and a determination to explore how to help others with disabilities. This drove me to want to focus on Biotechnology to create better futures...." Something like that.

I would say, just answer the prompt as much as possible. If it tells you to write about whatever, then write about whatever you feel is most significant to you, whether it regards your major or not.

Your essay topic sounds great.

The essay is an opportunity for you to tell the schools something about yourself that may not be obvious from the rest of your application. They will already know that you want to major in Biotech, and it sounds like your courses and ECs demonstrate your interest in that. So writing the essay about your interest in Biotech is not going to tell them anything new about you. Using the essay to show more about your character and personal growth is a great idea.
